    Hello,
i tried changing my ads positions and also used the 250* 250 square ads.
    Would this affect my earnings?

Yes it will. :) It can be in +ve or go -ve too.
If it goes good keep it or else try something else.
And yes you do need good amount of traffic first of all.

300*250, This is the best size. You should keep this size

Yes, changing ad position will definately affect your earnings. The only way for you to see the different is by doing trial and error. Most experts suggested to use ad that blend well with the site (color, size, location). Large rectangle and medium rectangle are the best performing ads.

I dont think change in position is going to affect your adsense earnings. Earnings depend on the traffic and keyword you are getting and clicks you are getting

    The bigger the better!
The 300*250 will get you more CPM Ads because it is web standard ad format so more advertisers use it
and 336x280 will get you a higher CTR because bigger Ads get clicked more often.

Also track each single Ad to know which performs best and then work on improving each ad.
