I would use winnipegsecondhand.com as your main domain name for the website, and just redirect winnipeg2ndhand.com to the main site.
Whilst both make sense (IMO) I would recommend that you use www.winnipegsecondhand.com instead of www.winnipeg2ndhand.com. Whilst www.winnipegsecondhand.com is 18 characters and thus 3 characters longer it does look better than www.winnipeg2ndhand.com. I guess that you own both so it might be a good idea to have www.winnipeg2ndhand.com redirect to www.winnipegsecondhand.com. Alternately have you considered rearranging different words such as "used", "preowned", etc for the domain name? Regards.
I think people like whole words rather then things like "2nd". It's easier to remember and way easier to read. But both domains make sense and as above poster said you could just direct both of them to the same place.

If you are going to have a classified site then you need a Content Management System (CMS), which will take a bit of time to set up but once working you can easily add, remove, and update content. There are loads of CMS out there, the main ones being: WordPress, Drupal, and Joomla. Once you have decided on your CMS you can look for addons that will help turn your site into a classified site.
I've had to Google "classipress", which came back that classipress is by AppThemes and is claimed to be "the most popular and widely used classified ads software for WordPress in the world today". I don't use WordPress and I don't know classipress. I suggest that you look/enquire in the DP WordPress forum.
Buying both and re-directing one to the other would protect your brand identity. I would definitely recommend using the first one as your site name, that is the "spelling" that most people will use as a matter of course. Also when you tell potential advertisers your site name you won't have to add "2nd, like the number two"
acutally, when i google winnipeg 2nd hand, i got both winnipeg second hand and winnipeg used, but no winnipeg 2nd hand, so, winnipegsecondhand wins out and usedwinnipeg dot com benefits in this game as well, no doubt!
