On page optimization is how you set up the actual pages of your site using H1,H2,H3 Tags, how you use bold, italics, underscore and of course how you use keywords on your page. Nowadays don't overdo your main keyword. Have it in say H1...but only once. Then say have it once more(at most twice) in the rest of the page. Look for around 1-2% keyword density at most otherwise you might get hit by Google. Off page optimization is how you go about getting backlinks to your site. Try and get only around 10-20% of your backlinks to be anchor text using your main keyword. This is especially true for exact match domains. Try and vary the other anchor text to be a mixture of related phrases to your main keyword as well as text such as "click here" etc.
