It depends on how much work you're willing to do afterwards. There is an argument that you're much better off submitting to 25 directories a week over 5 month instead of 500 all at the same time.
Search engines may be a lot less likely to discount a large proportion of links gained at the same time.

At the end of the day, nobody knows EXACTLY how Google, Yahoo and MSN interperet things apart from Google, Yahoo and MSN.
However - if you do things that are more likely to look as if links have build up naturaly then it's my opinion that you'd be better off.

It really depends on what you're using your website for. If you need loads of visitors straight away you might be better off to go for the 500 straight away option. But weight up your reasons and the pros and cons carefully.

Given that PR is a measure of link-popularity, getting links from directories isn't going to lower PR. But since not all links are counted towards PR equally, some links (I'd say: most directory links) will not help towards PR.

But the real deal we are all after is rankings in google. And crappy links to your site may hurt your site if you have no quality links to your site. For a new site I'd rather see only one on topic link from a website meant for humans, then 100 links from crappy directories.
