I am using two different computers to edit my dreamweaver website throughout the day. What is the best way to transfer this between computers and not have to deal with reconfiguring where my files are located each time?

Simply , Use Flash memory and keep it with you

Anther easy way is to use GmailFS (Gmail hard disk) , So everwhere you have internet + installed GmailFS driver , you could access your files
Myself i will pick GmailFS over usb stick , but its your choice :)
