Ive seen a few sites that use commas in URLs - definitely not spam sites.

Dunno why they do it. I wouldnt worry, but wouldnt use it on my own sites...

What would a "," possibly have to do with spam?

I've honestly never seen a comma in the URL. Not sure how they manage to do that.

I think it's a certain content management system that generates URLs with commas in them. I've seen them often at large content sites. For example http://www.thesun.co.uk/

Using comma separates 2 words, and saves space. Otherwise you will need to use 2 spaces, & or and. Sometimes I use dash (-) to separate 2 words instead of using a space.
