Which is best for SEO purposes?:

    domain/country1/region1
    domain/country1/region1/

    domain/country1/region1/hotel1
    domain/country1/region1/hotel1/

What are your thoughts on this , whether forward slash "/" is must present at the end of the urls

If region1 and hotel1 is a folder then forward slash "/" is must, so that search engines could give importance to all the pages in that particular folder.

it depends on what you are optimising for. As a rule of thumb, the further away from the domain name, the less relevant the content. However, the more specific your URL, the better for traffic.
