It depends on the rest of the content on your site. If you're creating a page on keyword1 that lists lots of other pages on your web page with associated content like keyword2 then I'd go for the keyword1/keyword2 format. Otherwise go for keyword1-keyword2 but I wouldn't duplicate any of the words.
I think the second one will be given more weightage than first one. Just wait for some more time to get more replies from DP members.
If the keyword1 looks more like a category containing keyword2 then do first one. At the end you are developing sites mainly for users. What looks natural to users should look valuabl to search engines now or later. But other than that situation, i would prefer the second way, since if you go one level deeper that most probably means your PR go to one level lower..
I would go for the second...the first puts keyword two one level deeper and you should try to keep your pages as close to the root as possible
This same thing was discussed in a different thread. I have tested both of these methods and I have gotten the URL with all the words together ranked into good positions. So I say all words together. Url 1!

Is best, and that's not an opinion, that's what I've found to be true in both Yahoo and Google through my own tests. But I'd only use it if http://www.mydomain.com/keyword1/ is also a valid page.
