I have a tumblr blog having 4000 Active Real followers, i mean when i post image they start rebloging it. Now my question is that, if i put my other website link in caption (description), and post the photo then people starts rebloging the photo with link, then the backlinks will be counted? for example if 40 peoples reblog the image with link in decription, then will i get 40 backlnks? And second question is that will they effictive or my domain PR?
You should install the Firefox plugin called "SearchStatus". If you did... then you will find a blue button on the bottom of Firefox. Right click and activate "Hightlight Nofollow Links". Then in the future all links which are nofollow, will be highlighted and all dofollow links will be normal. Good method to see if a link does count or not.

Your question is slightly confusing. Am I correct in thinking you have a Tumblr blog and a website and that putting a backlink from your Tumblr post to your website (not Tumblr) counts as a backlink? This will count as a backlink, whether or not it's dofollow or nofollow I don't know but you can edit this in the theme code. You can also automatically put backlinks into every post without you having to do anything extra other than create your post.

I'm pretty sure it will count for something, as Google gives more authority to websites that people are talking about on social media sites.. so when people are sharing and reblogging a post with your link in it, then it must be beneficial. Either way, I would imagine it helps with traffic? Not sure if you'll see every Tumblr sub-domain from users who have reblogged it show up on your Google Webmaster Tools links..
You need to be careful regardless of where you place your links whether on Tumblr or elsewhere. Randomize your efforts and don't overdo it.
