first of all - design depends on your niche, not 100% , but it depends. I mean if you have site with css3 tuts and all modern internet coding stuff - I wouldn't care about optimizing that site for ie6 - ie7, my visitors are supposed to be modern and geeky enough. 2-3 clicks to get to content Optimized look for mobile devices 960+ width or 100% (let's say fixed or 100%) - doesn't make difference, but depends on site, if it's a forum / portal / big e-shop - so do it 100%, if it's just a site (company, small store, whatever) - do it fixed
