I have the same problem I have a web site with flash online games, it is more then 1 year old and has been growing up very quickly last months... this site has been located in Top10 with some good niched keywords for some month, and 3 days ago they disappeared from google.com, but they still present on their places in other data centers such as google.ca, google.co.in, google.co.uk, google.com.au, etc... then I found it on 150-250-350 positions in google.com ... some other good keywords and related searches still present in TOP10 even in Google.com data center... Very interesting situation, specially with data centers... google.com threw away the TOP100, and others don't... what could it be?
You have to remember that Google makes changes that better the results for USERS. In no way is Google going to cater to the needs of webmasters as far as search results are concerned.
I've been freaking out about this all day. It is really weird. Some of the sites replacing top ranks are really crappy sites. I mean really crappy. I think google has gone NUTS
Same has happened here. But about every hour or two I move back up a placement or so. My luck they will keep climbing until they reach the top and then fall off and have to start all over again.
Google gives the new sites a small bosot for a little while then it gives them normal prority maybe thats why
This happens for many people, quite often. You just need to wait it out and weather that storm and you'll come out of the other side alot better off.
I experienced the same thing to some of my sites too around the same time frame as the thread starter. I guess the only thing to do is to keep building more back links, add more content, and hope for the best.
What is the best way to check backlinks? I know for sure that I have at least 200 new backlinks in the past month but google still reports 33 total for my site in Webmaster tools.

So, I hope that after Google finishes developing its new secret project we will get our lost positions where they used to be.
and here what I found about my opinion on Matt Cutts blog: Q: How do I check out the Caffeine update? A: If you search on www2.sandbox.google.com you can get a preview of how the search results will change over the next few weeks and months. so, here it is! If that is really so, we can believe that that our lost positions will be back!
Have lost SERPS major on a couple of sites but others have stayed the same or climbed ! Could it be Google is looking for more up to date content. Fresh content ranking higher?

I also had a similar experience, it was very scary! but after 10 days or so my SERP was regained! so give it a few days and you should be alright
