I want to get trademark for my current online+offline website & software development business.
I will explain here with an examples instead of revealing my original brand name.

e.g; I own "JKLSPY" Company... now what if someone own company with only word "JKL". they also trademarked "JKL" name.
Now can i trademark my "JKLSPY" name ??

PS: If i simply register my brand trademark, then is it applicable over the world ? or just for my region ? How much it costs in india to get trademark for brand name ?

That is not an accurate statement of the law.

There is not a problem as long as JKL and JKLSPY are unrelated companies that are doing different things and the name is just a coincidence. If your domain is an attempt to capitalize off of their brand (ie. cocacolatshirts.com, buyfacebookfans.com) there will be problems.

I am not a lawyer, and this is based on my understanding of US trademark and domain laws, it may be different in India.

Generally trademarks only function in the area or jurisdiction they're used in, so just in your
region. Check with your local trademark office on the costs and process involved.
