Whats the effect of using a different domain for a site?

I have done alot of SEO for site however I would like to if possible

However, I also have the domain in .com form with and without hyphens.

At the moment all the other domains use a 301 redirect to the hyphenated domain, however I have decided that I want to drop and market the site without the hyphen in the domain name.

Is doing a simple 301 directly effective to transfer all the SEO I have done for the hyphenated domain to a non hyphenated domain or either a .com domain instead of the .co.uk

    Ie. this-site.co.uk to thissite.com

Oh boy. If this site is targeted towards UK traffic, it makes more sense to use .co.uk. I personally don't like making people type hyphens, so I prefer to not have them in the URL.

BUT changing things will put your site back in the serps for at least a few months. Once the most used domain is a few years old, I'd personally just stick with that one. You might want to market the URL without the hyphens, just to have people avoid typing them.

Yeah, I was always going to use .co.uk anyway, I just wanted to get a picture of the whole idea.

Essentially the whole aim is to go from this-site.co.uk to thissite.co.uk but Im not sure if a 301 will be good for this task, sinceim already on position 2 for the best keyword in the industry it could be problematic to test.

Well if you do move, 301 IS the way to go. Personally I would advise my clients against it though if they are already ranking. They can use the version without the hyphen on their paper correspondence and stuff. I'd limit it to that.
