Is it going to hurt my site with search engines if I have a video that runs completely from left to right and 4 to 5 inches of monitor space down from top to bottom of the video with text the rest of the page.

the search engines spiders just read the source code so even though the video will cover a lot of the screen, the code wont be too extensive.

as long as you have enough other content to go in the page aswell you'll be fine.

Also, it doesn't really matter if you have a webpage that is not SEO friendly, that can't hurt the rest of the site. If that page is not SEO friendly it won't rank, but the rest of the site will be safe.

You can optimize it.. by separating it to another page and optimize by using meta-tags descriptions and meta-tag title etc....:D

using css helps by keeping the content seperate from design, this way all the search engine spider sees is pure semantic content and not a bunch of nasty table cells chucked in for design purposes. code to content ratio plays a part so its always good to have nice clean code.
