Depends of SE algorithm. SE algorithm might change in the future. I would optimize site for both flower and flowers, since it is also varied anchor text .
It absolutely makes a difference. With the LSI algo running currently it does make sense to optimize for keywords, their plurals and typo's. However, optimizing for all of those things can be an energy drain, which is why your initial keyword analysis is crucial.

You would be ranked better if your 1. Title has both "flower" and "flowers" 2. Description has both "flower" and "flowers" 3. your page name has "flower" in it. Having said that, "flower" is a broad term and people always search with "some specific term" + flower. My suggestion would be to target for specifics.
As far as I have seen some similar cases, if you continue optimizing for flowers, you will get traffic even if people search for flower.
