I would like to know how one does run a blog.site with subdomains. blogspot.com and wordpress.com would come to mind here. But I've seen that other people are doing this as well.

Let's assume I've got a site myblog.co.za / This would be the main entry page. and also the page where people have to sign up and login.

If they are signing up, they can create a new blog for them. like forinstance

    subdomainname.myblog.co.za

They'll have their own control panel to run their own blog.
Would there be software available to run this? And of course what would be the requirements towards the hosting server?

yes you'll need Wordpress MU, but you can't have a sub-sub domain!

WPMU is the one and only, I think :).
try WP Webhost, they're dedicated to wordpress hosting, they have WPMU hosting package option.
