Optimizing a website for long tail keywords is relatively easy. Just ensure that the keyword is near the beginning of your title and somewhere in your content. For a more competitive long tail, you might also need to include that phrase in your headers, alt text and/or meta tags. A more competitive long tail will also require some backlinks to that page using your keyword as link anchor text.

The bigger challenge will long tail keywords is actually finding good long tail keywords that get enough searches, but aren't too competitive. To really do well with long tails, you might want to invest in <a href="http://www.keyword-research.info/paid-keyword-research-tools">paid keyword research tools</a>. These will give you more detailed stats for low volume keywords.

You can also use web2.0 site to optimize for these long-tail keywords. they seem to rank pretty high for these long-term keywords particularly squidoo and hubpages.

Well, including your targeted long tail keywords in your title and header tags would greatly help your site to rank. I suggest you target long-tailed search terms that have searches and are less competitive. In that way, you can easily rank on them and at the same time receive great amount of traffic.
