Well Im working on a niche directory now after I wanted to run a general for the last 8 months or so. Niche is definitely easier and can be more profitable than a general one, however there really isnt anyway way to know for sure. Just concentrate on building a quality directory which ever type you choose and you should be fine.
This month I made on a general directory with adsense $25 this month a niche directory with paid inclusion $1000+

When making a directory you've gotta think about users. If user like it, it will get popular and lots of traffic. This will intern bring you paid submissions.
I think it's pretty late to start new directories, too many webmasters run directories, and not just one...some have several
Do people click on ads on directories. I would have thought people only really click on content sites.
Hehe. One year ago when I was starting, I remember to have read an article that "just about everyone has her/his own directory already". One year later.... hundreds or thousands more were started Directories have generally very low CTR. Most directories' only traffic is search engine robots, webmasters submitting, and webmasters checking for their links via search engines, there are in fact only very few directories that have some REAL traffic worth of discussion.
Can you promote a general web directory? If you have lots of sites on different topics, you may have a good outlet for promoting a general directory. If you have only sports sites, you might go sports directory (for example).
