Any tips of how ad-spot pricing should be decided?

    e.g.

Someone wants the leaderboard space on one of my sites.

(Currently the site has minimal ads).

It gets around 40000 uniques per month.

How much should I ask for the leaderboard space?

Depends on the specific site, but most advertisers prefer CPC.

On average 1 in 100 pageviews gets a banner click. So if you have 40,000 uniques that would be somewhere around 100,000 - 150,000 pageviews per month, which would work out to 1,000 to 1,500 clicks per month on average.

Based on the links in your signature, I'm going to guess this is for your casino site. It's not uncommon to charge $1-$5 per click for gambling ads, sometimes more if you have high quality traffic.

So I would contact a few online casinos and ask for their internet marketing department and work out a deal with them. I could provide you with the contact information for a big one if you want.

Best bet is to build a media kit before doing advertising though, or at least advertising page on your website at the bare minimum.

If you want some help PM, I've done this hundreds of times with various websites.
