You are doing it backwards. Find you keywords FIRST. Then build your site around those keywords. Simple...
I think maybe some keyword tools can help you much. For example, Google adword keyword selector, it's an excellent tool of getting keyword suggestion.
Be sure to start with Twitter...this will help you see what's most popular. After that, run the results through the Google Wonderwheel and you'll be on the right path.
Can you elaborate more on how to do keyword research in Twitter? Never heard of anyone using twitter as a keyword research tool.
Sure Ashique, Unfortunately, I have to start off by stating that one of the greatest info-filters on Twitter is now off-limits, or as they say - Forbidden. It was discovered that by adding a dash to your keyword (example -wordpress) you could get hyper-accurate results of the keyword sepcifically as it was used. Doesn't mean twitter isn't still useful. As Zeeksteen pointed out, you should always find your keywords before creating a product and Twitter is a great filter. Start by running your keywords through the Google wonderwheel, then after determining which ones are most tightly correlated, join them together in Twitter search to see how frequently they come up in conversation.
Ok.. I get it, but I'm not sure how effective Twitter can be to research your market. From my personal experience, I've only seen marketers talking about (or promoting or even spamming about) their products/services in twitter. If I want to figure out what my chosen market (ideally my 'prospects') are talking about my product/services that I (want to) offer, I'd try to examine relevant forums, blogs, and yahoo answers to do so. In twitter, non-marketers usually talk about random stuffs happening in and around their lives. But when they've a question to ask (on anything related to a product/service that we offer), they either 'Google it' or go to relevant discussion forums to find the solution. Having said that, I also realize twitter can be a tool to measure how many competitors you have in your niche who are constantly promoting similar products and services. But then again, you can get more accurate results for measuring you competition through Google's Keyword Research tool.
I suggest you should go with Google Adword Keyword Research Tool. It will give you all the information about particular keyword:- a) How Competitive Keyword it is. b) Monthly Local Search c) Monthly Searched all over the world on that particular keyword.

just like it was pointed out the best way to start a fresh site is to do a diligent research on what is appealing to people at the present moment. ie what are people talking about, you can use google trend to check what is hot in the market. you can also use google search box by typing action words like "how/what to buy" "how/what to purchase" etc. this is the first step when buying a domain name from your search you can now build your site around the keyword phrase
