    Well, I think some things would depend on your niche.

    I would say:

    Do have a good landing page - The way your page looks is vital to weather someone will give you their email and want to be a part of your list.

    You can share poll and pollers must fill name and email address to put vote. Using Public Surveys can also help in generating huge list of Emails.

    Building a list is important because you can market to those people for free over and over again.

    Do's: Get with a reputable company like aweber or getreponse.
    Dont's: Don't email everyday.

    Well its pretty easy, If you have a linkedin account then join a group named as 500 Plus+ Link: http://www.linkedin.com/groups/500-Plus-3792456. People shared their email addresses in this group it can be more then a 100,000 email addresses. I think its the best way to build email list..
