Does anyone have ideas on how I should go about getting people to talk about my site on Twitter? I'll be launching the blog after I reg the domain and I see Twitter as a valuable tool for promotion. Any ideas?

You might want to check out Sponsored Tweets. You can pay to have your site talked about on Twitter by users with the follower count you need to promote.
Follow other social media heads on Twitter, RT their post and if you have quality links, they'll RT your post as well.
Depending on blog content, you could use hashtag that might already be in use for similar subjects. That way it turns up for anyone following that hashtag.
i love the way moonfruit used twitter to promote there 10th birthday i was amazing, pure marketing genious! they gave away 10 macbooks for 10 days to people who #moonfruit in there tweets. people posting saying stuff like... "I love the smell of #moonfruit in the morning" and .. "I'll #moonfruit you if you #moonfruit me" , "#moonfruit its all about luvin" sparked tweets like "Why is everyone talking about #moonfruit?" baseiccly... it was top tweeted about topic for them 10 days
I think unless ur site is really big or u have a good marketing strategy (with a good budget), twitter won't really work for u in the short run. but, if u add a follow me link on ur blog and gradually keep adding followers and keep tweeting u can get something out of twitter.
True, I'm not sure how beneficial Twitter would be to me because my site is already a blog...not sure what exactly I would tweet.
Twitter has been more than valuable for more and more people and so using it effectively should be your focus. You can get carried away at times.
