It definitely works, just look at some of the more successful forums on the internet, how many do you see that doesn't allow signature? Especially important if it's a forum targeting webmasters or in the IM niche.
Talk to the people who actually pay for DP members to use their link they provide the user with. If it didn't do anything Im sure they wouldn't be paying for it.
Allowing forum signatures will increase site popularity and keep people coming back, It also helps keep the discussion going. I have come across allot of dead or dieing forums and those tend not to use sig links.

The fact that You can have a signature link will make a lot of people participate more and of course they will try to deliver good content so people become interested enough to click on the link. You will have an ocasional spammer but it's not something to worry i supose. It dependes on the niche but usually people are responsible. Start by allowing 1 sig link. See how it goes. I've seen forums going down because of spammers but I don't think that should stop You from trying. DP allows sig links and there's nothing going wrong!
I think that allowing people to place a link in signature have only good sites: if someone would like to post only for link when adding it into signature he brings more content to the forum and don't spam it in fact. Of course people are sometimes only posting because of those links but it's a great chance that after a while it will be just a regular member. It's worth giving a try!
