You wouldn't want to make a blog from the main URL and then later change it. You'd end up with a bunch of pages that can no longer be indexed once you switch to a store front.

If the blog is something that you'd keep going, then it would be worth making it as mystore.com/blog.

I'd say to start building the content and general design of the site over the next couple months.

DO NOT submit your site to any search engines, including any sitemaps, until you are satisfied with it so far.

keep your domain clean by all means!!!
and forget the idea with the blog.

there is no way to start seo before your online store is life except you have (a) related business webpage(s) from where to drive traffic to that store.
this is what I did, as my company site was online many years before.
a single static page would do the job as it is out of google cash within days.
I would not want to have dozens of pages in the index that don't work later on.

now some additional personal advice if you want any. :)

do all the beta testing on a local server and go online when your software is bug free and working.
make sure the shop software does not pass session IDs to the robots!!!!!!

once the business is online, the linkbuilding will have to start.
what you will need are links from old ( trusted ) domains.
no matter what their PR is, they will give you the strongest boost.

directories, bookmarks and all that stuff do work of course but the results are slow and the linkpower in most cases is zero.
instead of using one of the cheap submission services here on DP I would suggest to submit your site manually yourself using unique descriptions to the few strong directories and pay for inclusion in the even fewer top directories out there. you may use those services to get some extra links of course.

for the rest...search the forums...there is more information than you cn handle. :)
