I use SEOtool bar and on there they have a little NF button that you just press to check if the links are do follow or not. If you use firefox there is loads of add on as well.
A more tedious alternative would be to just right click on a link and select properties. With Firefox if the link is a nofollow it will show nofollow in the link properties box if its do follow it will not show nofollow anywhere in the link properties box
Firefox users are fortunate enough to have a NoDofollow add on. The add on can basically confirm if a link if dofollow or nofollow. You can google this addon or you can check out my signature to checkout a tutorial on how to install it.

Just open the source code, locate that link you want to check and confirm if there is rel="nofollow" or rel='something else nofollow" within anchor tag. These firefox plug-ins do a great job too.

Just to be sure, even with my SEO program, I usually check the source code for the nofollow tag. When you combine that with the Find function (Cntrl + F), it does not take long at all.
Definitely check the source, you might be surprised at how many modules manage to hide the nofollow attribute. I noticed this with IntenseDebate - the right-click properties trick didn't show nofollow, but the source had it in there.
