Option 1, IMLTHO, would be best. I have used some online tools for checking my keywords and found that some of them would get confused if there was not a space in additon to a comma. I presume, I don't know for certain, that some search engine bots could have the same problem. As for the keywords you are proposing to use, there is a lot of repetition in your keywords. Some search engines will only read your keywords as 'real estate new homes sale ca sc santee'. You might want to consider using different keywords rather than repeating the same keyword over and over.
Use comma to separate keywords. That's meta keyword and by that name it refers to its contents as keywords and not phrases composed of keywords. If it does not take too much of your time, you can still put it there though its impact is minimal at best.
