Can this be done? Regards to Craigslist.

    Make an ad that ask visitors to reply to my craigslist post. Once the user replies to the post it triggers my email to send them a email with my Affiliate Ad in it?

    Sure it can be done with Gmail's autoresponder.

    In your ad just write something along the lines of "For more information reply to this thread" which then sends you an e-mail and in turn Gmail automatically sends your autoresponder message. Just make sure to set it for a few minutes delay to make it seem more natural.
