Lately I have been reading that any font emphasis doesn't help Google rankings. I personally still think it would help rankings on Yahoo and MSN, but probably not so much on Google. If it does help, it probably isn't significant. These days backlinks tend to trump any onpage factors.

No but <h> tags do; it helps outline the parts of your content (heading, sub-heading, etc.). Not just by appearance though. They are recognized by search engines.

I don't think bold, etc. helps in Google any more. Not sure about the other search engines.
H1, H2, H3 tags tend to help a lot more than any font emphasis...

there are six heading tag size h1, h2, h3, h4, h5, h6. The top heading tag of ur pages should be wrapped in h1 tag
ad should add ur page main keyword and keypharses

Bottom line - after you've set a basic hierarchy using title and heading tags, organize, write and style your content so it's appealing to your users. This will pay much greater dividends in the long run (user retention) than trying to finagle things around to please search engines. In the end, if it's a well organized document, and your users like it, chances are search engines will too - since their goal is also to help searchers find the most meaningful and well organized content.

You can use <h>,<b>,<i>. These are considered in google also. But I don't think mentioning size isn't mater.

Even if it doesn't help per se with search engine rankings, remember that it WILL help actual humans who view your site! Bolding/italicizing important parts of your site/article will definitely help the readability of your document.

Just make sure it isn't done obnoxiously!

Instead of using bold/italic font, try internal linking. Say you want to target the homepage, go to a different page of your site include your homepage keywords somewhere in that content. Then make these keywords phrases a link to your homepage.
