Can Wordpress handle and be used for big business portal site that has e commerce transactions, database profiles of few hundred thousand businesses, messenging/ chat , forum community etc.? Or is it better to start from scratch and do native ? Thanks

Mag themes normally have integrated BB Forums plugin to manage active community, chatroom, db profiles etc. too. For example, Green Mag wordpress theme has the functions you required.
