Community is everything with podcasting (I have been a podcast producer since 2005). Hang out at places such as podcastalley.com and make connections with other podcast producers. Get them to play your promotional audio/video spots, link to you, and talk about you and your podcast on theirs. Other podcasters' existing audiences are the quickest way to build your audience at the start. Don't forget standard web promotion such as search engines, SEO, etc, as that's vital, too of course.
