Is there like any formula or a general rule of thumb regarding how much backlinks should a new site receive. Later on it could be adjusted as the site grows and becomes bigger and older. Should there be more Web2.0s, Wikis, Article Submissions, Social Bookmarking, PRs. I believe these are the type of backlinks you would want for a quality Tier 1 but I am not sure how much of them should be build and for what period of time. I do not want to overdo it and look spammy.
i can't say the actual number even if you post 10 per day. google bot wont crawl all of them. for me i aged my domain for a month or two, add some content and do on page seo. i'm not even submitting it to google webmaster. i need to also decide first what will be my sitemap.xml format.
It's difficult to calculate a number or determine how much you need exactly. Unfortunately, link building is not an exact science. Much of it is trial and error. What you can do is build links slowly, building more and more each day. You can start with 10 from each method for now and then build on that each month. I think it might vary depending on the niche you are in. Also if you can build up other trust factors you will probably get more leeway. So make sure you get some high quality links, link out to some trusted sites and regularly post unique content. Technically a site should be able to receive unlimited new links if it proves it is trustworthy. Even things like listing an address and a privacy policy might help. Look into Trust Ranking.
If you are consistent in adding backlinks everyday then it shouldn't matter the number. You just have to make it look "natural" to google. Getting a bot to do it is probably not the best way to go about getting backlinks in the beginning...or ever.

Also, Google loves variety, meaning, they should contain assorted links: Web 2.0, Forum, Social Networks, Article Directories,etc. Got the gist? Otherwise the links built will become less worthwhile.
No there cannot be one. Sometimes only 5 links from good and trusted websites overpower a 100 links from low PR websites. SO this formula cannot possibly be drawn.

You want maybe 50 to 100 HIGH QUALITY backlinks on .EDU sites. After the site has been around for a few months, you can create more. Google doesn't like it when brand new sites get tons of backlinks, and they penalize for it.

Now you need social signals coming from your site also but you need to build same amount of links consistently and you will win the race, don't forget you need high PR relevant theme links that super important and also a diversity of links from all kinds of sources.
