you can use your keywords on a Title (little Sentence that include the 2 keywords with no need for "," ==> transparency)

no problem but i'm just suggesting you not to use if you don't want to do but there's also no trouble to use comma for Bots

You can use comma to separate between 2 keywords.
For example: "social networking site" will be read by bots as a combination of 3 words but "Social Networking, Social Bookmarking" will be read as 2 combinations of 2 words. So yes, you can use comma. It depends on what you what the bots to read.

Title as in <title> in the <HEAD> section or a page's <h1> tag?

You can use it, why would you not be able to use it, however if you wanted to gain any value then using characters like that are of no real use, EXCEPT for readability for humans.

yeah, comma really needed, it makes some sense and differienciate two keywords from each other in Title.
May or may not be it recommended for se's but for real human visitors, it required to have comma btwn one or more keywords.
