Yes of course it could! Although you have to realize that a single page website can only be optimized for so much keywords. If you want a broader set of keywords you should add more pages with different types of content related to your main keyword.

Therefore every page has its own ranking. How well these pages rank individually has a large effect on your website’s overall ranking. Also if your website is penalized then you will receive lower rankings for your individual pages within your site also.
Although individual pages are counted for ranking keywords, overall backlink profile to your website is also considered as major factor for ranking well in Google.
