Get an Askimet account for starters. It's also helpful to add no follows to the link or not allow link entrees at all. Take away the benefit of posting spam to your site and that is a good start. In the end you will always get a few tho.

I have found disqus to competently remove spam from comments, personally I love it as a commenting system as a whole but you might like to try getting it and turning it on for your blog.

Requires a bit of work but may well be worth it for you, check out www.disqus.com. Oh and just as a note it is essentially an entirely separately hosted commenting system (free) but you can export and import if you ever want to turn it off and fall back to just the plain old Wordpress commenting system.

Akismet is by default already a plugin in WordPress. Use that as it's really good.

What I found effective is Spam Free Wordpressplugin which is free and kills spam 100%. I used Akismet and some still pass through. The plugin's author also has another plugin that kills trackback links.

Use a combination of akismet and approve all blog comments manually. Akismet will pick up the majority and the rest are easy enough to spot.

Signup for an API key. Been a while since I set one up, but I think it does actually tell you where to get one while setting it up.

askimet does not have rules to catch spammers it just looks in it's database if spammer is in there database they put it in spam folder. It does not detect spam on it's own. I read about a plugin Bot Blocker never used it though maybe you can check that out

Try to use WP-Optimize as well. It's a good tool to clear spams as well.

Install Raven's Antispam - it is the most effective antispam plugin which prevents 99 % of bots from posting comments.
