I know some large sites like "about dot com" use sub domains with a lot os success. I have seen others do it as well, for topics completely unrelated. It exploits google's weakness (a catch 22 in their ranking algo)
It all depends how often spiders index your site and the site that you post your links on. Example: If you are making blog comments with your keyword subdomain and that blog is pretty dead (it hasn't been updated or indexed by robots frequently) then it will take long for the search engines to recognize your backlinks and give your site credit.
