What do you want the site to do ?

Are you selling products, blogging, managing a forum ?

There is a ton of different sites out there, what do you want ?

Anyway, to proceed you need hosting. After that you need to install software or get someone to write it for you and then get people to visit your site.

Decide what you want to do.

If you don't know html or have a good web design program, start with a blog. Wordpress or Blogger are pretty easy to install and use and maintain. There are even many Wordpress templates that don't even look all that "blog like" if you want more of a web feel. But like fulltilt said, you need to decide what you want to do before you decide anything else.

You can also set up a free blog on blogger.com and then map your domain to it.
That will get you started without hosting.
    eg. http://www.indiemall.com/

If blogging I would use a all in one CMS

Like wordpress or Joomla, you can find all sorts of professional templates online for them free.

its simple as others are saying.. get a hosting. maybe ur domain registrar also gives u hosting.
most of the hosting companies use cpanel. u can configure ur site from there.
but the main is the html file which u need to upload through a ftp software like cuteftp.
or you can also install scripts like joomla, wordpress etc. through fantastico in cpanel.

Look through wiki articles for web hosting and web design. Now you need to have web design for your web site and then you will need to host it.
