.com is the best for return visitors because it is easily remembered. Other than that, it really doesn't matter.
.com is the best like money moose said, but .net and .org is good too.. Find an example joomla site.. If you have a chance to choose, then just choose .com (if you develop any general site), but if it don't, you can choose .net.. And if you are building any network site, you better use .net. If you build an organization site, you can use .org,, business with .biz

I would go for a .com first, but as so many of these are bought up and parked waiting for a buyer, I would then go for a .net if your .com name is not available. I've never seen the need to buy a .biz domain name...
Mixsp... I have said this many times... My personal & professional belief is that .com shall always rule. Reason: We are conditioned to type in .com as an extension. When is the last time you typed in .net, .biz, .info etc ??? No, please don't. .biz is useless - name me a popular website with a .biz domain? Most people just automatically add .com to the end of a domain name, so if your website is AcmeInc.biz a whole bunch of people will type in AcmeInc.com by mistake.
Usually, .org's are for non profit websites. Like American Cancer Society, etc. .com's are used for everything. Businesses, fan sites, help sites, etc. .Net's are usually used for more network sites (I'm guessing it'd be ideal for IRC and chat websites). And I can't really say for what .biz's are used for because more businesses use .com

In all technicality an extension is just a period followed by two or three letters. However, like Kyle said, there is generally a convention as to how they're used. The common user really doesn't think about this. However, when it comes to the consumer, it's always a safe bet to use .net or .com. Most people follow convention due to the popularity of the phrases that surround .com when you're referring to business. Besides, like a few people have mentioned, a ton of people tend to recall .com before any other common extension.
