There are other threads out there as well - I suggest you read those as well. Just bear in mind that real on-site SEO optimization is done before you write one line of code or one word of Web copy on your page.

Before you even think to build links or something you need to make a good and detailed research for the keywords you are going to target (i.e. the keywords you want people to find you from search engines). Once you have made the list of the keywords you intend to target you can start building your website already, and use the keywords while you building your site, in the title of the site, in the h1 tag, images title and alt tags, use anchor text during the internal linking, make search engine friendly (or so called) URL's (i.e your-key-word.htm). I have wrote a first part article on Building your website thinking SEO, this article is mainly talking on what you are asking. I had no time to go to the second step, but i will soon. In poor words, you need to think about insite seo before you even dare to start working on offsite seo, main things you need to remember: Title tag, h1 tags, anchor text for internal site linking, image title and alt tag and a clean code.
also..i would defintly say start with keyword research, find the most profitable low competition keywords and then go from there. SEO optimize your site for the keywords you find.
And while you're optimizing for "the long tail" don't forget to optimize for those competetive keywords you want to target as well. Most of it's the product of good copywriting, so if you can write killer Web copy (which includes both those major and minor keywords) you're halfway there. If you don't know how to do that, I strongly recommend you read "Web Copy That Sells" by Maria Veloso. It should be available at your local bookstore or public library (if the library doesn't have it, they should be able to get it via an interlibrary loan for you).
