All main keywords must be in Title, domain name, header tags, meta tags, and anywhere in the content to top SERPs.
I agree with boyponga's post and would like to add that with regards to content it is no longer effective just to rely on putting your keywords in the article. It is now better to use LSI methods to get a closely themed content.
LSI means Latent Semantic Indexing. It's a technology that some people think G is using to extract the meaning of the content from a page, even if the exact words or search phrases do not exist on the page. Whether or not G is using LSI is a debatable topic. If you are building strong themes for your pages, you are probably focusing on the the elements that have a positive impact with LSI.
Best tip for SEO that I have found (at least for google) is to get backlinks with the appropriate keyword anchor texts. You can rank for any keyword term if you get enough backlinks with that term as part of the anchor text. For this to appear natural you have to vary the actual keyword phrase used as the anchor text, but always keep your main term as a part of the anchor text phrase.
well its hard to think of some when there arent any if we knew the "new" methods we would all be rich because we would be first I predict we dont see anything new in this thread but maybe new people will get some ideas from it
Google may be using LSI to a limited extent in current serps, but it also appears that they are using more phrase related ranking information. (relational data based on phrases, phrase relevance, and phrase density compared to the corpus as a whole.
How about filling my meta tags with "britney spears, pamela anderson, britney spears, pamela anderson, britney spears, pamela anderson, britney spears, pamela anderson, britney spears, pamela anderson, britney spears, pamela anderson" Surely that's got to work! I've got another KILLER strategy too... Why not have LOTS of HIDDEN TEXT exactly the same color as the background!!!!

I've seen different suggestions listed before but what is best practice for varying anchor text. I've been using three different iterations for example in directory submissions. Several people mentioned it in this thread but I'm curious as to what most of you out there are doing that seems effective?
In LSI, Google ranks website in SERPs based on how strong the overall THEME (Site Flavor) is of a website for the searched keywords against as % of keyword optimization for a site currently. Try using tilde "~" in front of a keyword in Google to see the difference yourself. ~keyword
