Moving Site....301 Redirect ok?

    I have a site - mysite.com and a directory for it - mysite.com/directory
    mysite.com used to have an article dir on it but I got rid of it and now it is just one page.

    My mysite.com/directory has just got a pr4 and I want to reopen it and do away with the mysite.com page.How can I do this without being penalised?would a 301 redirect in htaccess do it ie redirecting mysite.com to mysite.com/directory or is there a better way?
