Is it good practice to do SEO for one keyword phrase and when it's ranking is "good" jump for another keyword OR do SEO optimization for all my keywords at once? I have in mind on- and off-site optimization.
From my experience you are as well to stick to a few keywords , and concentrate as off site seo allows you to work with lots of keywords , but onsite is a little bit more difficult if you want more than a few.. So I would go for one and whens its up where you want work from there... Best of Luck in your efforts!

one by one is not very good approach ... this is length procedure because search engine updates your rankings in a week or more !!! .... so you have to wait longer to see the results. Instead better, target some set of keywords (keywords set should be smaller so you can track them easily) and you can have better analysis on SERP's. Have good time with SEO
2 or 3 would be better, for example keyword: MLB Baseball Store its can be combine between: MLB Store, MLB Baseball or Baseball Store its my experience.
In my experience of performing Search Engine Optimization for my clients, and myself, dependent upon the targeted keywords/phrases, I would typically recommend you target at least three terms at once, agreeing with the statements given above. The best approach to optimizing your site for three terms or less would again be to focus on the on-page optimization of the site; From there, I "may" skip back links and focus more on traffic-generating link exchanges it they are strategically planned out.
Do long tail first..this way you can focus on a 5-8 terms...move on to the next bunch and then hit big...
Start off with 5 or 6 keywords that are not popular and therefore easier to rank for. Then work your way up.
At first you can't really rank on one word keyword. But try to concentrate first on 3-4 word keyword.
This depends on 1.) What keyword you are targetting. How competitive it is. 2.) Your experience @ SEO and your budget.
Design your home page to be targeted to your main key-phrase and maybe another sub-page for that main phrase as well but DIFFERENT content on each. Design the rest of your pages each for its own keyword with articles, resources and information. They will begin to rank especially if you target long-tail phrases. 2 visitors a day per phrase X 100 keywords is 200 visitors a day and you will probably never have to worry about losing them. Don't do spam type pages that are useless but write good content.
You may want to target more than 1 keywords...guess 3 major keywords and another 3 minor keywords...as targeting 1 keyword at a time will takes you a lot of time...i agree with what Visio told us...you may want to do what he'd told...
I'd recommend selecting keywords you will be using from the beginning, 1000's of links with one anchor text looks shady. Use multiple keyphrases....
