In my opinion, link building with related websites and link directory websites, is more effective than link building with forum signatures, article submission, and press releases. Keep in Mind: Every website is different! Some methods may be more effective for some website and less effective for other sites. Every website should be considered on an individual basis.
related sites are not only better, its a must. if you cannoot get related sites, then get them created. when you do article submission, build web 2.0 pages and mini blogs, they provide related contextual backlinks. dump the forum signatures as they are almost useless in terms of backlinks. well, they do show up as backlinks but hardly have any value
link building to relevant sites is a great help but don't forget that working on other strategies like article submission, pres release will also help your website.
True related websites will count a lot. Although it would also count if you post your sites' links in related forums and in their related categories in web or article directories.
Its not a question of this or that is better, the fundamental principle to guide you by is 'the more links, the better'. But google also calculates the value of the incoming link in terms of the pagerank of the incoming link.
A lot of forums include no-follow tags on posts to discourage people from posting just to get their post count up. It's also not too useful to have backlinks from forums if the particular thread has no PR (and threads take a long time to get PR bled to them from the main domain). I'd say definitely go with the related sites.

Does forum signatures harm websites in any way or if my site get hundreds of backlinks from a single domain, say I submit many articles on a single Article Directory, or many forums signatures, Is there any bad impact?
Usually no. If your website is new, it could get a penalty which expires sooner or later anyway. It is my own experience; am just having one website suffering a penalty because it had got a sitewide link (which doesn't exist any longer now). On the other hand, if your website has got hundreds or thousands of links, and if it's relatively old (say over 1 year), then thousands of links coming from a single domain won't hurt it's rankings.
Link building with the related sites is better.if you have the backlinks in the related sites it helps to improve the ranking of the site.But anyway you have good traffic if your site have more backlinks.
