I am using same as OP at all my wp blogs.

    ilfilosofo.com/blog/wp-db-backup/

It works really well (weekly scheduled backup).

I use WP-DBManager by Lesterchan: http://lesterchan.net/portfolio/php/ - It not only allows scheduled backups but also allow you to restore to older backups, optimise DB and even perform sql queries(not the DROP ones ofcourse).

Make sure the pathname is correctly setup. CHeck the Main DBManager options and please post the exact error. ANd check this post for some solutions: http://forums.lesterchan.net/index.php?topic=3714.0 - Read full post for some alternative solutions. And do type in the url of backup-db folder to correctly check that it should not open.

There is no harm in manually but suppose you missed it by any chance and at the same time something happen with your blog then ?.. Better you go with scheduled wp backup option using any good plugin IMO for your own safety ;)

If you and your host both not having your DB backup then you can't do anything. You have to start everything from the beginning. Always good to install WP backup plugin.
