Does it matter whether you have no-follow or do-follow? Or only that the link should be related to your niche. To submit to quality directories, I think you should have quality content, traffic and backlinks. The sites which are new will not get submitted on these directories as they lack quality content, etc. So You should submit to both kind pf directories; one which are new and others which have reputation. Most of the edu links are no-follow. The how can they be a help to a person? Actually you have to maintain a balance between all the factors PR,traffic,content,etc. If you have high pr and no traffic then what is the use of getting a pr. If you have content and no traffic then again no profit from the blog. And if you have traffic and no content, you will loose bounce back rate and slowly the traffic. I think try to keep everything equal and you will succeed in what you want to achieve!
Yeah, IMO the content is one's body, and the traffic is blood, Pr is face(i do think it not so ipmport for a profit aimed website, i looking for it just for the face)!
Don't be too concerned with PR unless you're selling links - focus on your search engine rankings instead. Getting dofollow backlinks from high PageRank authority sites should increase it though - obviously if the page your link is on has pagerank it will be worth even more

Try to create backlinks on high pr sites! it can really effect to develop good PR! Some times! Quality Links on selected sites can impress your Site PR
