In this case, I don't think you're referring to the TLD. .NET is a software framework (think: programming languages) developed and promoted by Microsoft. I personally don't mess with it but believe it's mainly used for developing Windows applications.
Nitroshock is right and, yes, it only works with Windows software so it is limiting to some degree. Some will say it works on *nix with Mono but no self-respecting *nix developer or user would install Mono.

No. As mentioned above, .NET is a framework that interfaces between languages and Windows. It's advantage is that one can code in multiple languages and they can still compile and run on Windows at the same time (being very simplistic here). Something that has been easy to do on *nix forevery. Not directly but, nowadays, a Windows programmer would probably use the framework for interoperability reasons. Has nothing to do with web sites ending in '.net'. The internet runs on Unix.
