If you cannot get the .com your after, what would be your next choice i.e. .net or .org etc...
Also, are there any disadvantages to choosing another domain name extension?
Thanks..

Definately .net. It's worth a lot more than a .org.

Compared with the value of the name, yes there are disadvantages the non .com TLD will worth less
However it all depends on you, if and how you can establish traffic to your other TLDs and brand your site
My second choice would be .net and then .info

it depends on the name. I only register a .net if it has a potential. my second choice is .org if it is related to an organization but if not i won't buy.

If the name is important i.e. for your business, register all of the extensions that you can afford.
