I currently have a website that has a PR5.

If I change the theme of the site completely (change the subject of the site), will the PR be affected?

No. It will remain as long as the backlinks to it remain.

It depends on nature of your backlinks if they are natural then no doubt they will remove your link when they will see you are no longer relevant to their contents but if you have bought backlinks then there will be no change :)

though it would be like starting battle for keywords from scratch in Google :)

page rank is a number which is calculated with formulas and the variables of this formula is backlinks . not content of the site . if you keep your backlinks , your page rank will not change

Increase your PR There are several ways to Increase :-- directory submission, article submission, RSS syndication, reciprocal linking, blogs, forums...

As long as your backlinks remain and the PR update is already finish your PR will not change.
